Pengaruh Ekstrak Etanol Bit Merah (Beta vulgaris L.) terhadap Jumlah Arteriole Endometrium pada Tikus (Rattus norvegicus) yang dipapar Asap Rokok

Paparan asap rokok sering terjadi dikalangan masyarakat, ini dapat menyebabkan terjadinya peningkatan ROS sehingga antioksidan dalam tubuh menurun. ROS yang meningkat akan menyebabkan gangguan pada organ reproduksi seperti endometrium dan ovarium. Akibat gangguan ini maka akan berpengaruh pada tingkat kesuburan (infertilitas) pada wanita. Bit merah mengandung senyawa betalain yang tinggi akan antioksidan yang dapat menetralisir ROS yang ada didalam tubuh yang dapat mengganggu organ reproduksi wanita yang dapat menyebabkan salah penyebab ketidaksuburan pada wanita. Penelitian ini bertujuan untuk mengetahui pengaruh ekstrak etanol bit merah (Beta vulgaris L.) dengan berbagai dosis terhadap jumlah arteriole pada endometrium tikus yang dipapar asap rokok. Jenis penelitian ini adalah true experiments dengan rancangan post test only control group design. Menggunakan 25 ekor tikus betina yang dibagi menjadi 5 kelompok yaitu kelompok kontrol negatif (tanpa perlakuan), kelompok kontrol positif (tikus yang dipapar asap rokok 2 batang/hari) selama 56 hari, kelompok perlakuan I (dipapar asap rokok 2 batang/hari dan diberi ekstrak etanol bit merah dengan dosis 125 mg/kgBB/hari) selama 56 hari, perlakuan II (dipapar asap rokok 2 batang/hari dan diberi ekstrak etanol bit merah dengan dosis 250 mg/kgBB/hari) selama 56 hari dan perlakuan III (dipapar asap rokok 2 batang/hari dan diberi ekstrak etanol bit merah dengan dosis 500 mg/kgBB/hari) selama 56 hari. Metode yang digunakan untuk melihat jumlah arteriole pada endometrium tikus dengan menggunakan pewarnaan Haematoksilin Eosin. Kemudian masing-masing slide diamati dibawah mikroskop Manual Dot Slide Olympus XC 10 yang dilihat per lapang pandang. Data dianalisis dengan menggunakan One Way Anova, hasil nilai p-value jumlah arteriole (p=0.000). Hasil penelitian membuktikan bahwa pemberian ekstrak etanol bit merah dengan dosis 500 mg/kgBB/hari dapat meningkatkan jumlah arteriole pada endometrium tikus yang dipapar asap rokok. Penelitian ini membuktikan bahwa pemberian ekstrak etanol bit merah dapat meningkatkan jumlah arteriole pada endometrium tikus yang dipapar asap rokok. Kesimpulan pada penelitian ini adalah terjadi peningkatan jumlah arteriole pada kelompok yang diberi ekstrak etanol bit merah dengan berbagai dosis. Tetapi dosis ekstrak etanol bit merah yang dianggap paling cepat meningkatkan jumlah arteriole adalah kelompok perlakuan III dengan dosis 500 mg/kgBB/hari.

Exposure to cigarette smoke often occurs among the community, this can cause an increase in ROS so that antioxidants in the body decrease. Increased ROS will cause disturbances in reproductive organs such as the endometrium and ovaries. As a result of this disorder it will affect the level of fertility (infertility) in women. Red beets contain betalain compounds which are high in antioxidants which can neutralize ROS in the body which can interfere with the female reproductive organs which can cause one of the causes of infertility in women. This study aims to determine the effect of red beet (Beta vulgaris L.) ethanol extract with various doses on the number of arterioles in the endometrium of rats exposed to cigarette smoke. This type of research is true experiments with a post test only control group design. Using 25 female rats divided into 5 groups namely negative control group (no treatment), positive control group (rats exposed to cigarette smoke 2 cigarettes/day) for 56 days, treatment group I (exposed to cigarette smoke 2 cigarettes/day and given red beet ethanol extract at a dose of 125 mg/kgBW/day) for 56 days, II treatment (exposed to cigarette smoke 2 sticks/day and given red beet ethanol extract at a dose of 250 mg/kgBW/day) for 56 days and treatment III (exposed to cigarette smoke 2 sticks/day and given red beet ethanol extract at a dose of 500 mg/kg/day) for 56 days. The method used to see the number of arterioles in the rat endometrium using Haematoxylin Eosin staining. Then each slide was observed under a Manual Dot Slide Olympus XC 10 microscope which was viewed per field of view. Data were analyzed using One Way Anova, the results of the p-value of the number of arterioles (p=0.000). The results of the study proved that administration of red beetroot ethanol extract at a dose of 500 mg/kgBW/day could increase the number of arterioles in the endometrium of rats exposed to cigarette smoke. This study proves that administration of red beetroot ethanol extract can increase the number of arterioles in the endometrium of rats exposed to cigarette smoke. The conclusion in this study was that there was an increase in the number of arterioles in the group given red beet ethanol extract at various doses. But the dose of red beet ethanol extract that was considered the fastest to increase the number of arterioles was the treatment group III at a dose of 500 mg/kg/day.

IV. PROOFS
Galley proofs (PDF format) for an accepted article will be sent by e-mail to the corresponding author for copyediting corrections. The core content of an article cannot be changed at this time. V. Â REPRINTS
Five free reprint copies of the published article will be shipped to the corresponding authors in Indonesia only, approximately two weeks after the issue is printed. Authors, including overseas authors, who wish to purchase reprints, are required to complete the Reprint Order Form, which will be sent with the galley proofs to corresponding authors. This form must be returned together with the corrected galley proofs.
