Abstrak Latar Belakang: Indonesia memiliki kekayaan biodiversitas, termasuk tanaman herbal seperti daun Lawsonia inermis Linn. Biasa di kenal dengan daun pacar kuku, dikenal dalam pengobatan tradisional sifat antibakterinya. Namun, penelitian ilmiah tentang efektivitas daun pacar kuku terhadap bakteri patogen, seperti Propionibacterium acnes dan Escherichia coli, masih terbatas. Bakteri ini dapat menyebabkan infeksi kulit dan pencernaan, yang sering diobati dengan antibiotik, tetapi penggunaan antibiotik berlebihan menyebabkan resistensi. Penelitian ini mengeksplorasi kandungan fitokimia ekstrak etanol daun pacar kuku dalam mengatasi infeksi bakteri sebagai alternatif pengobatan berbasis herbal. Tujuan penelitian ini untuk mengetahui kandungan fitokimia dan aktivitas antibakteri parameter Kosentrasi Hambat Minimum (KHM) dari esktrak etanol daun pacar kuku pada variasi kosentrasi terhadap Propionibakterium acnes dan Escherichia coli. Metode penelitian yang digunakan ekperimental laboratorium, . Uji KHM ekstrak daun pacar kuku menggunakan metode dilusi cair kemudian diukur kekeruhan dengan spektrofotometer UV-Vis. Konsentrasi estrak yang digunakan 5%, 30%, 45%, 60%, 85%, ,100% serta Kontrol positif (tetrasiklin 30µg) dan Kontrol negatif DMSO (1%). Hasil penelitian ini menunjukkan ekstrak etanol daun pacar kuku (Lawsonia inermis Linn.) memiliki kemampuan menghambat bakteri Propionibacterium acnes dengan rerata zona 18-24.85mm, sedangkan Escherichia coli rerata zona sekitar 6.21-11.78mm. Nilai KHM pada kosentrasi 6.25-50%. Disimpulkan pada penelitian ini ekstrak daun pacar kuku (Lawsonia inermis Linn.) mempunyai aktivitas antibakteri terhadap Propionibacterium acnes dan Escherichia coli memiliki kemampuan menghambat (KHM) Saran: Ekstrak daun pacar kuku ini berpotensi dikembangkan sebagai sediaan topikal seperti krim atau gel untuk pengobatan jerawat, dan uji klinis diperlukan untuk memastikan keamanan dan efektivitasnya. Commonly known as henna leaves, known in traditional medicine for its antibacterial properties. However, scientific research on the effectiveness of henna leaves against pathogenic bacteria, such as Propionibacterium acnes and Escherichia coli, is still limited. These bacteria can cause skin and digestive infections, which are often treated with antibiotics, but excessive use of antibiotics causes resistance. This study explores the phytochemical content of henna leaf ethanol extract in treating bacterial infections as an alternative herbal-based treatment. The purpose of this study was to determine the phytochemical content and antibacterial activity of the Minimum Inhibitory Concentration (MIC) parameter of henna leaf ethanol extract at various concentrations against Propionibacterium acnes and Escherichia coli. The research method used was a laboratory experiment. The MIC test of henna leaf extract used a liquid dilution method and then measured the turbidity with a UV-Vis spectrophotometer. The extract concentrations used were 5%, 30%, 45%, 60%, 85%, 100%, as well as positive control (tetracycline 30µg) and negative control DMSO (1%). The results of this study indicate that the ethanol extract of henna leaves (Lawsonia inermis Linn.) has the ability to inhibit Propionibacterium acnes bacteria with an average zone of 18-24.85mm, while Escherichia coli has an average zone of around 6.21-11.78mm. The MIC value at a concentration of 6.25-50%. It was concluded in this study that henna leaf extract (Lawsonia inermis Linn.) has antibacterial activity against Propionibacterium acnes and Escherichia coli has the ability to inhibit (MIC). Suggestion: This henna leaf extract has the potential to be developed as a topical preparation such as a cream or gel for the treatment of acne, and clinical trials are needed to ensure its safety and effectiveness. Further research is needed to isolate and identify the active compounds that play a role in antibacterial activity. III. EDITORIAL REVIEW AND REVISION

The reviewers' (and editor's) suggestions will be conveyed to the author, who will then have an opportunity to revise the paper. A manuscript returned to an author for revision can be held for a maximum of eight weeks.

IV. PROOFS
Galley proofs (PDF format) for an accepted article will be sent by e-mail to the corresponding author for copyediting corrections. The core content of an article cannot be changed at this time. It is the author's responsibility to check the entire manuscript, including tables, figure legends, and cited reference numbers, not just the items queried. The proofs should be corrected and mailed to the editorial office within 48 hours of receipt in order to expedite publication. Only one set of corrections will be accepted. V. Â REPRINTS
Five free reprint copies of the published article will be shipped to the corresponding authors in Indonesia only, approximately two weeks after the issue is printed. Authors, including overseas authors, who wish to purchase reprints, are required to complete the Reprint Order Form, which will be sent with the galley proofs to corresponding authors. This form must be returned together with the corrected galley proofs.
